I opbygning og stil minder Johannespassionen i høj grad om samtidens oratoriekompositioner, for eksempel Händels "Messias". Men oratoriegenren udvikles i Johannespassionen til sit fuldeste med dens rollefordeling der svarer til de medvirkende i historien, således at handelingen fremstår med en operas dramatik. Operaens sceniske og dramatiske aspekter får i passionen og oratoriet en anden ramme at udfolde sig i: I stedet for teaterscenen træder nu alteret og kirkens rum ind som "kulisse" for det musikalske drama lidelseshistorien er.
Evangelisten synges efter traditionen af en tenor, og Jesus og Pilatus af to basser. Også andre mindre roller er fordelt på solister. Koret agerer både er vrede jøder, hånende soldater og repræsenterer den lyttendes refleksion over lidelseshistorien igennem alle koralerne og de smukke arier.
I barokkens Tyskland var det almindeligt at opføre musikalske passioner ved gudstjenesterne i den stille uge op til påske. Derfor er de tre opførelser af Christian IV’s Vokalensemble også bevidst planlagt i netop disse dage i ugen op til påskedag. Johannespassionen er mere end blot en koncert, men i høj grad et stykke liturgisk musik, skabt til refleksion og fordybelse.
